What it is

Collagen is the most abundant structural protein in the body, forming a major component of skin, tendons, ligaments, cartilage, and bone. Hydrolysed collagen (collagen peptides) is collagen that has been enzymatically broken down into small, more readily absorbed peptides, usually sourced from bovine, porcine, or marine (fish) tissue. The body's own collagen production slows with age, which has driven interest in collagen supplements for skin appearance, joint comfort, and nail health. Evidence is strongest and most consistent for reducing activity-related joint pain in physically active people. Evidence for skin hydration and elasticity is positive when pooling all available trials, but that overall effect appears heavily influenced by industry funding and study quality rather than being a robust, independent finding. Evidence for nail growth is weaker still, resting mainly on small, uncontrolled studies. Collagen peptides are generally regarded as safe and well tolerated.

    Hydrolysed collagen is widely marketed to improve skin hydration, elasticity, and wrinkles; pooled trial data show a positive effect, but this appears to depend heavily on industry funding and study quality rather than being a robust, independent finding.

    A 2025 meta-analysis of 23 randomised controlled trials (1,474 participants) found that, pooled across all studies, collagen supplements significantly improved skin hydration, elasticity, and wrinkles. However, in subgroup analysis by funding source, studies without pharmaceutical industry funding showed no significant effect on any of these outcomes, while industry-funded studies did; similarly, high-quality studies showed no significant effect while lower-quality studies did. The authors concluded there is currently no clinical evidence to support using collagen supplements to prevent or treat skin ageing.

    Collagen peptide supplementation (around 5 g/day) may modestly reduce activity-related joint pain in young, physically active adults with exercise-related knee discomfort.

    A 2021 randomised, placebo-controlled trial in 180 physically active adults (18-30 years) with exercise-related knee pain found that 5 g/day of specific collagen peptides for 12 weeks produced a significantly greater reduction in exercise-induced knee pain than placebo (-21.9 mm vs -15.6 mm on a 100 mm visual analogue scale, p = 0.024), confirmed by physician assessment (p = 0.003); pain under resting conditions did not differ significantly between groups.

    Weak, preliminary evidence from a small uncontrolled study suggests collagen peptides may increase nail growth rate and reduce brittle-nail symptoms, but this has not been confirmed in a placebo-controlled trial.

    A 2017 open-label study with no placebo control, in 25 women with brittle nail syndrome, found that 2.5 g/day of specific bioactive collagen peptides for 24 weeks was associated with a 12% increase in nail growth rate and a 42% decrease in broken-nail frequency, with 64% of participants rated as clinically improved. Because the study had no control group, was small, and was affiliated with the collagen manufacturer's research institute, this result should be considered preliminary rather than confirmatory.

    Hydrolysed collagen is generally well tolerated in clinical trials; studies of up to 6 months have reported no significant adverse events, though most trials are relatively short and small, so long-term safety data remain limited.

    A 2025 randomised, placebo-controlled trial of low-molecular-weight collagen peptides (3,000 mg/day for 180 days) in 80 adults with knee osteoarthritis reported no adverse events in either the treatment or placebo group over the 6-month study period, consistent with the generally favourable tolerability profile reported across other collagen peptide trials.

Dosage & how to take it

  • Hydrolysed collagen peptides: 2.5–15 g/day, any time of day, with or without food. Trials showing skin or joint benefits typically used 2.5-15 g/day for at least 8-12 weeks; effects, where present, are gradual rather than immediate. Lower doses (2.5-5 g/day) are typical for skin/nail use; higher doses (10-15 g/day) have been used in joint-focused trials.
